General Introduction of Nanobody Customization Services

Nanobodies, or single-domain antibodies, originate from the VHH region of camelid heavy-chain antibodies such as those in alpacas. At only about 15 kDa, roughly one-tenth the size of conventional antibodies, they feature a simple structure, strong stability, and high affinity, making them widely useful in tumor immunotherapy and cell-based therapies.

Stage Service Content TAT Deliverables
Stage 1: Antigen Preparation (Optional) * Protein antigen purity and endotoxin identification (>5mg/protein antigen)
* Protein antigen conjugation to carrier protein followed by immunization (Optional)
2-4 weeks Protein Antigen Identification Report
200ug of antigen conjugated to carrier protein
Stage 2: Alpaca Immunization & PBMC Isolation * 1 adult immunized alpaca, immunized with two protein antigens 5+ times, 0.5-1mg each time
* PBMC Isolation
8-10 weeks ELISA report for post-3rd immunization serum
Stage 3: Construction of VHH Phage Display Antibody Library * RNA extraction and cDNA preparation
* Amplification of VHH gene fragments, construction of nanobody immune library
* Library quality control: Pick 96 single clones for sequencing, analyze sequence diversity and correctness
4-6 weeks Intermediate products (e.g., RNA or cDNA); Provide library QC report, VHH library size of 108–109;Phage Display Nanobody Immune Library (library phage particles 10ug)
Stage 4: Phage Display Panning & Clone Identification  3-5 rounds of panning using antigen pressure
* Pick at least 288 clones for ELISA validation
* Identify and evaluate positive clones
* Sequencing of positive clones and analysis of sequencing results
4-6 weeks Eluate from the last round of panning; ELISA screening report; Provide 3-5 unique sequences per protein antigen
Stage 5: Nanobody Expression, Production & Validation * Clone 3-5 nanobody genes into expression vector
* Recombinant antibody expression and SDS-PAGE validation
2-4 weeks 0.1-0.5mg purified nanobody (SDS-PAGE >90%, endotoxin level < 1EU/mg) and corresponding strain; Nanobody ELISA test report

Note: The requirements of antigen provided by customers: purity >85%, concentration > 1 mg/mL, PBS buffer.
