Monoclonal antibody uses:
● Quantification of protein expression level
● Immunocytochemistry (ICC), immunohistochemistry (IHC), and immunofluorescence (IF)
● Detection of structural changes and of antigens' phosphorylation states
Polyclonal antibody uses:
● Capture of as many antigens as possible (e.g. IP or CHIP)
● Detection of denatured proteins
● Detection of proteins which may have undergone glycosylation or morphologic changes
Advantages：
• Highly specific recognition of only one epitope of an antigen
• hybridoma cell lines have the ability to produce unlimited quantities of antibodies
• High consistency
• Minimal background noise and cross-reactivity
• Excellent for affinity purification
Disadvantages：
• More expensive
• Longer production time
• Possible lower signal
• More vulnerable to loss of epitope through chemical treatment of the antigen. This can be offset by pooling two or more monoclonal antibodies
