The cells of different samples were infected with viruses carrying CRISPR-Cas9 sgRNA library. The cells were then sequenced with next-generation sequencing (NGS) technology.

We first evaluated the effects of sgRNA knockout and compared the enriched and depleted sgRNAs; the genes and targets were then screened for cell survival or drug action through functional annotation. Gene set enrichment analysis involves finding overexpressed classes in a group of genes or proteins; the most common forms of such analysis are GO annotations and KEGG pathway enrichment analysis. Using gene pathway enrichment analysis, we were able to conduct a preliminary analysis on the biological processes or signaling pathways that differentially expressed genes may participate in. Finally, the targets were further analyzed by combining the cancer database and mutation database.
