When choosing an RNA polymerase, select the appropriate enzyme according to the promoter sequence on the DNA template, e.g. T7 RNA polymerase is suitable for T7 promoter, T3 RNA polymerase is suitable for T3 promoter, and SP6 RNA polymerase is suitable for SP6 promoter.
The template for RNA synthesis is a strand of DNA that is transcribed by T7, T3, or SP6 RNA phage polymerase in the presence of ribonucleoside triphosphates (rNTPs). This polymerase moves along the template strand, using base pairing with the DNA to create a complementary RNA strand, substituting uracil for thymine.
RNA oligos should be stored at -20°C or lower in RNase-free water or specialized RNA storage solutions to prevent degradation. Avoid repeated freeze-thaw cycles during storage to maintain their integrity.
