shRNA Structure Sequencing

shRNA design is typically divided into two formats: the simple stem-loop shRNA and the microRNA-adapted shRNA (Fig.1). Due to the hairpin structure, shRNA is difficult to sequence in gene synthesis. Universal sequencing protocols can hardly work, especially for the second structure’s sequence (Fig.2). However, Synbio Technologies has developed and enhanced the protocol, and obtained high quality sequencing results (Fig.3).
